Book excerpt: By monitoring the surface induced dissociation (SID) products as a function of increasing collision energy or from sub-complexes initially produced from SID of the native complexes (in SID-IM-SID experiments), we were able to gain a fundamental mechanistic insight into the assembly of several tetrameric protein complexes. In addition, the SID collision energies associated with appearance of the SID fragments may be used as a means of characterizing the relative strengths of the inter-subunit contacts. The coupling of IM with collision-induced dissociation (CID) and SID allowed us to identify and characterize the structures of the conformational intermediates present during the unfolding of several complexes, and thus illustrates its capability to probe the folding mechanisms of protein complexes.
